How It Works

Double-stranded DNA functions mechanically as a semiflexible polymer with high torsional stiffness. In topologically constrained genomic domains, the fundamental topological linking number (Lk) is partitioned between double-helical twist (Tw) and tertiary superhelical writhe (Wr). Torsional stress generated during transcription and replication induces plectonemic supercoils that regulate gene expression.
